NOW L-Proline 500mg, 120 Capsules

L-proline is one of the amino acids collagen is built from, and these capsules help support the body’s own collagen production for skin, joints and healing tissue.

Roughly a quarter of the amino acids in a collagen strand are proline or its hydroxylated form, so it is a fair ingredient to reach for. The part almost every seller leaves out is that proline cannot become collagen without vitamin C, and that if you are eating protein at all you are unlikely to be short of proline in the first place. Both facts change what you should buy.

Proline Without Vitamin C Does Not Become Collagen

This is the single most useful thing to know before buying any collagen ingredient. Once proline is built into a collagen strand, an enzyme has to convert part of it into hydroxyproline, and that enzyme cannot work without vitamin C. No vitamin C, no conversion, and the collagen that results is unstable and falls apart.

That is not a theory. It is the mechanism of scurvy: sailors with plenty of protein and no fresh fruit developed bleeding gums, wounds that reopened and skin that broke down, because their bodies had all the proline they needed and no way to use it.

So take this with vitamin C, or with food that contains it. A guava, an orange, a handful of raw kale or a vitamin C tablet alongside costs very little and is what makes the proline usable. Buying proline and skipping vitamin C is buying half a reaction.

Who Is Likely to Benefit, and Who Is Not

Recovering from surgery, a burn or a significant wound. This is the clearest case, and it is why proline appears in clinical nutrition for wound healing.

Eating very little protein, through illness, appetite loss or a restricted diet.

Everybody else, honestly, is less likely to notice much. If you eat meat, fish, eggs, beans or dairy most days, you are already taking in far more proline than these capsules provide, and the thing limiting your collagen is more likely vitamin C, total protein or plain ageing. A collagen peptide powder, which supplies proline alongside glycine and the rest of the sequence, is usually a better buy for skin and joints than a single amino acid.

What It Will Not Do

It will not remove wrinkles. Skin ageing is driven mostly by sun exposure and time, and no amino acid reverses that — daily sunscreen does more for the appearance of skin than anything in this aisle.

It will not treat arthritis or repair worn cartilage. It supplies a raw material; it does not direct where the body uses it.

And it is not a treatment for heart disease or blood pressure. There is an old theory that proline and lysine with high-dose vitamin C clear arteries. It has never been supported by controlled trials, and anyone with heart disease or high blood pressure should follow the treatment their doctor has prescribed.

How to Take It

Two capsules, once or twice a day, preferably on an empty stomach where amino acids absorb best. That is 1,000mg to 2,000mg a day, and 120 capsules gives thirty to sixty days depending on which you choose.

Take vitamin C at the same time. If healing is the reason, give it the length of the healing rather than a fixed period, and make sure overall protein intake is adequate too, because collagen needs the whole set of amino acids and not only this one.

Before You Start, Check These

Speak to our pharmacist first if any of these apply:

  • Kidney disease — amino acid supplements add to the nitrogen the kidneys must clear.
  • Liver disease — the same principle applies.
  • Pregnancy or breastfeeding — there is not enough information on supplemental doses.
  • Any other amino acid supplement — they compete for the same absorption routes, so taking several at once can make each less effective.
  • A restricted or medically supervised diet — worth checking that a single amino acid is the right addition.

Side Effects

Proline is well tolerated and most people notice nothing. Mild nausea or stomach discomfort occasionally occurs and usually settles if you take the capsules with a little food, at a small cost to absorption.

Stop and speak to us about a rash, swelling of the lips or face, or persistent stomach pain.

Proline, Collagen Powder or Vitamin C?

If your reason is skin or joints and you are eating normally, a collagen peptide powder gives you proline in the company of the other amino acids collagen needs, and there is more human research behind it. If your reason is healing after surgery or a wound, proline alongside adequate protein and vitamin C is a reasonable, targeted choice. And if you are not already taking vitamin C, that is the cheapest and most certain part of this whole equation.

Our pharmacist will work out which of the three fits your situation, including when the answer is that food and a vitamin C tablet will do.

Storing It

Keep the bottle closed, dry and out of direct sun, away from bathroom and kitchen steam. Use it within the date on the label and keep it out of reach of children. For adults only.

Frequently Asked Questions

What is L-proline used for?

It is one of the two most abundant amino acids in collagen, so it is taken to support the body’s own collagen for skin, joints, tendons and healing tissue. Its clearest use is recovery from surgery, burns or significant wounds, when demand for collagen rises sharply.

Do I need to take vitamin C with it?

Yes, and this matters more than the dose. The enzyme that converts proline into the hydroxyproline collagen needs cannot work without vitamin C. Without it the collagen produced is unstable, which is exactly what happens in scurvy. Take a vitamin C tablet or eat fruit alongside.

How many capsules a day?

Two capsules once or twice daily, preferably on an empty stomach, which is 1,000mg to 2,000mg. A bottle of 120 lasts thirty to sixty days.

Will it get rid of my wrinkles?

No. Skin ageing is driven mostly by sun exposure and time, and no amino acid reverses it. Daily sunscreen does more for how skin looks than anything else you can buy, and our pharmacist can show you what is worth using with it.

Is proline or collagen powder better for skin and joints?

For most people, collagen powder. It supplies proline together with glycine and the rest of the sequence, and there is more human research behind it. Proline on its own makes more sense when the goal is healing a wound rather than general skin and joint support.

Do I even need it if I eat normally?

Probably not. Meat, fish, eggs, dairy, beans, gelatin and bone broth all supply proline generously, and your body also makes its own. The usual limits on collagen are vitamin C, total protein and age rather than proline itself.

Does it help joint pain?

It supplies a raw material for cartilage but does not direct the body to repair a particular joint, and it is not a painkiller or an anti-inflammatory. Joint pain that persists for weeks deserves a proper assessment rather than a supplement.

Is it safe with kidney problems?

Ask first. Amino acid supplements add to the nitrogen load the kidneys clear, so anyone with kidney or liver disease should check before starting.

Can I take it with other amino acids?

You can, but they compete for the same absorption routes, so taking several at once can make each less effective. If you are already on another amino acid, bring the bottles to the counter and we will work out the sensible order.

How long before I notice anything?

Collagen turnover is slow, so think in months rather than weeks for skin or joints. For wound healing the relevant period is the healing itself. If nothing has changed after three months, proline is probably not what was limiting you.
